The ventricular system's accumulation of cerebrospinal fluid is a causative factor in the prevalent condition of congenital hydrocephalus. Currently identified as causally related to hydrocephalus, whether individually or as a widespread clinical sign, are four major genes: L1CAM, AP1S2, MPDZ, and CCDC88C. We describe three instances of congenital hydrocephalus, arising in two families, and attributed to bi-allelic mutations in the CRB2 gene. This gene, previously recognized for its role in nephrotic syndrome, is now further implicated in congenital hydrocephalus, a relationship characterized by some variability. Two instances of renal cysts were observed, contrasted with a single case of isolated hydrocephalus. Our neurohistopathological analysis demonstrated that, diverging from prior suggestions, the pathological mechanisms of hydrocephalus caused by CRB2 variations involve atresia of both the Sylvian aqueduct and the central canal, rather than stenosis. While CRB2 is known to be important in establishing apico-basal polarity, immunolabelling experiments in our fetal samples showed normal localization and expression levels of PAR complex components (PKC and PKC), tight junction protein (ZO-1), and adherens junction components (catenin and N-Cadherin). The act of disconnecting from the surrounding world, a phenomenon often referred to as mind-wandering, is a common experience that has been found to be associated with decreased cognitive performance in a variety of tasks. Within the framework of a web-based study, a continuous delayed estimation paradigm was used to examine the impact of task disengagement during encoding on subsequent location recall. Task disengagement was evaluated using thought probes, employing both a dichotomous scale (off-task versus on-task) and a continuous response scale (ranging from 0% to 100% on-task). This approach permitted us to contemplate perceptual decoupling, both dichotomously and gradationally. Our initial research, involving 54 participants, demonstrated a negative relationship between encoding task disengagement levels and the subsequent recollection of location, measured in angular degrees. This discovery favors a gradual perceptual disconnection process over a complete and instantaneous decoupling. Replicating the earlier result, our second study (n=104) demonstrated the same finding. A study of 22 participants, exhibiting adequate off-task activity, enabled the application of a standard mixture model. Using in vivo neuroimaging, we explored the effect of MB on cerebral blood flow (CBF) and brain metabolism in human and rat models. Two MB doses (0.5 and 1 mg/kg in humans, 2 and 4 mg/kg in rats), administered intravenously (IV), caused a decrease in global cerebral blood flow (CBF) across both species. This effect was statistically significant in humans (F(174, 1217) = 582, p = 0.002), and rats (F(15, 2604) = 2604, p = 0.00038). A considerable decline in human cerebral metabolic rate of oxygen (CMRO2) was found (F(126,884)=801, p=0.0016), accompanied by a substantial decrease in rat cerebral metabolic rate of glucose (CMRglu) (t=26(16), p=0.0018). This result casts doubt on our hypothesis that MB would increase CBF and energy metrics. Our results, remarkably, remained reproducible across various species, exhibiting a direct relationship with the dosage. Potentially, the concentrations, although clinically meaningful, exemplify the hormetic effects of MB, which implies higher concentrations leading to an inhibitory rather than an augmentative metabolic response.
